Buffer.<\/li><li>Antigen retrieval, pH 6.0 (times will vary according to microwave).<\/li><li>10 mM sodium citrate, pH 6.0, and heat in a microwave at 96<sup>o<\/sup>C.\u00a0*We usually do a series of three runs (6-7 minutes each run) to equal the time\/temp because of evaporation (refill coplin jars with dH<sub>2<\/sub>O).<\/li><li>Microwave according to instructions on microwave.<\/li><li>Cool on countertop, 15 min.<\/li><li>Rinse with dH<sub>2<\/sub>O<\/li><li>1X PBS, 5 min.<\/li><li>Block in 4% Goat serum\/PBS-T, 2 hours at RT.<\/li><li>For mouse anti \u2013<a blocking buffer, For guinea pig anti-SCGB1A1 (G210, Bl. 3\/24\/11) dilute 1:1500, for mouse anti-\u00a0<a href=\"http:\/\/www.sigmaaldrich.com\/catalog\/product\/sigma\/a5228?lang=en\u00aeion=US\" target=\"_Blank\"><strong>ACTA2<\/strong><\/a>\u00a0 (\u03b1 Smooth muscle actin) (<a href=\"http:\/\/www.sigmaaldrich.com\/catalog\/product\/sigma\/a5228?lang=en\u00aeion=US\" target=\"_Blank\"><strong>A5228<\/strong><\/a>, Sigma)\u00a0dilute 1:2000 in blocking buffer. Spin for 10 minutes in a \u00b5fuge, apply to tissue, and incubate on tissue overnight @ 4<sup>o<\/sup>C.<\/li><\/ol><p><strong>Day 2<\/strong><\/p><ol><li>Rinse slides in PBS-T 3X, 5 min.<\/li><li>Apply secondary antibody , Goat Alexa Fluor 488 anti-mouse IgG1 (<strong><a href=\"http:\/\/www.lifetechnologies.com\/order\/catalog\/product\/A21121?ICID=search-a21121\" target=\"_Blank\">A21121<\/a><\/strong>, Lot# 1345050, for anti \u2013<a href=\"http:\/\/www.sigmaaldrich.com\/catalog\/product\/sigma\/c7113?lang=en\u00aeion=US\" target=\"_Blank\"><strong>CALCA<\/strong><\/a> (CGRP) ) dilute 1:200, Goat Alexa Fluor 555 anti-guinea pig IgG (<a href=\"http:\/\/www.lifetechnologies.com\/order\/catalog\/product\/A21435?ICID=search-a21435\" target=\"_Blank\"><strong>A21435<\/strong><\/a>, Lot# 1571699, anti-SCGBA1A1) at 1:200, Donkey Alexa Fluor 647 anti-mouse IgG2A (<strong><a href=\"https:\/\/www.lifetechnologies.com\/order\/catalog\/product\/A21136?CID=search-a21136\" target=\"_Blank\">A21136<\/a><\/strong>, Lot# 1345055, <a href=\"http:\/\/www.sigmaaldrich.com\/catalog\/product\/sigma\/a5228?lang=en\u00aeion=US\" target=\"_Blank\"><strong>ACTA2<\/strong><\/a>) at 1:200, in blocking buffer. Vortex and \u00b5fuge at full speed for 10 minutes. Incubated at room temperature for 1 hour.<\/li><li>Rinse in PBS-T 3X, 5 min.<\/li><li>Dilute DAPI 1:2000 and apply to slides for 10 min.<\/li><li>Wash in PBS-T 3X, 5 min.<\/li><li>Rinse slides in 0.1M PB or TB, 3X, 5 min.<\/li><li>Add 1 drop of Prolong Gold anti-fade mounting medium (P36930).<\/li><li>Coverslip with Gold Seal Coverslip (Cat# 3422 Electron Microscopy Sciences, 22 X 22 mm).<\/li><li>Allow Prolong Gold to cure overnight at room temp in light sealed box.<\/li><li>Store slides in a light sealed box @
